They are obligatory blood sucker arthropods which economically impact cattle industry by reducing weight gain and production. Moreover, they are important vectors of viral, bacterial, rickettsial and parasitic pathogens infecting humans and animals. In view of the importance of Hyalomma anatolicum anatolicum in pathogen transmission, including Theileria lestoquardi in Iran, the accurate identification of this tick is critical. Although many keys are available as aids, morphological identification of tick species such as Hyalomma anatolicum anatolicum (Koch, 1844; Hoogstral and Kaiser, 1959) is difficult and expert knowledge is required. False morphological identification at the level of species and subspecies is common, particularly for Hyalomma excavatum complex members which are prevalent in Iran. For example, the high similarity between Hyalomma anatolicum anatolicum and Hyalomma anatolicum excavatum is the cause of confusion in the identification of these species. In this study, polymerase chain reaction (PCR) techniques were used for identification of Hyalomma anatolicum anatolicum based on analysis of the gene sequence of the Second Internal Transcribed Spacer (ITS2) of this tick. The ITS2 nucleotide sequence of Hyalomma anatolicum anatolicum was 963 base pairs (bp) in length and exhibited 93% homology with other GenBank registered ITS2 sequences of this subspecies (accession no: FJ593700.1). The complete ITS2 region sequence was identified in this study and registered in GenBank under accession number HQ123320.}, keywords = {ITS2}, title_fa = {بررسی توالی نوکلئوتیدی ITS2 کنه هیالوما آناتولیکوم آناتولیکوم در ایران}, abstract_fa = {کنه گروه مهمی از آکارین ها هستند که با هجوم به حیوانات و خونخواری، با ایجاد کاهش وزن و کاهش تولید باعث آسیب اقتصادی می شوند، بعلاوه ناقل پاتوژنهای مهم حیوانی و انسانیند. آنها به عنوان ناقل مهمی برای انتقال بسیاری از عوامل بیماری زای ویروسی، باکتریایی، انگلی و ریکتزیایی مطرح هستند. با توجه به نقش کنه هیالوما آناتولیکوم آناتولیکوم در انتقال عوامل پاتوژن از جمله تیلریا لستوکاردی در ایران، شناخت دقیق انگل این کنه بسیار مفید است. هر چند کلید تشخیص های فراوانی برای شناسایی کنه ها از جمله برای تشخیص گونه های هیالوما وجود دارد، اما استفاده از کلید های شناسایی مورفولوژیک خیلی آسان نیست و نیاز به افراد متخصص دارد. معمولاً اشتباه در تشخیص گونه ها وتحت گونه های کنه ها خصوصاً هیالوماهای گروه اکسکاواتوم که کنه های شایع در ایرانند به وفور اتفاق می افتد. به عنوان مثال، هیالوما آناتولیکوم آناتولیکوم بسیار شبیه هیالوما آناتولیکوم اکسکاواتوم است و این مساله باعث شده در تشخیص آنها دچار سردرگمی شویم. برای حل این مشکل، در این مطالعه از واکنش زنجیره ای پلیمراز (PCR) برای بررسی توالی نوکلئوتیدی ناحیه(ITS2) کنه هیالوما آناتولیکوم آناتولیکوم استفاده شده است. اندازه ژن ITS2 به دست آمده 963bp بود. توالی نوکلئوتیدی ITS2 هیالوما آناتولیکوم آناتولیکوم به دست آمده در این مطالعه با آنچه درGen Bank ثبت شده بود 93 درصد شباهت داشت.(Accession no FJ593700.1)}, keywords_fa = {}, url = {https://ijvm.ut.ac.ir/article_23103.html}, eprint = {https://ijvm.ut.ac.ir/article_23103_9f525a7bad3c7ff5f695e595aa63c5f1.pdf} }
