Effects of CH223191and Resveratrol on Bone Turnover Markers and Osteogenic Gene Expression in Osteoporotic Rats

Abstract
Background: The systemic metabolic condition of osteoporosis developed from failure in skeletal homeostasis, where accelerated bone resorption dominates over mineral deposition leading to impaired bone mineral density.
Objective: This study was conducted to assess whether the administration of resveratrol (RES) or the AhR antagonist CH223191 could mitigate the osteoporotic changes induced by ovariectomy (OVX), comparing individual treatments with a combined vas approach. Methodology: Fifty female rats were randomly distributed among five experimental groups. A Sham-operated control, an untreated OVX and those receiving resveratrol, CH223191, or combined therapy, throughout forty-five day experimental timeline, the systemic response was monitored via bone turnover, serum biochemical markers especially ALP, PTH, calcium, osteocalcin levels and focused on bone gene expression Tnsalp, Bglap. Integrated with a detailed histological analysis of the femur section to assess tissue integrity. Results: Data analysis revealed that OVX model exhibited marked elevations in bone metabolism indicators coupled with significant histological damage. Conversely intervention with RES, CH223191 or their combination effectively attenuated these pathological shifts and optimized bone gene activity.
Conclusion: RES and AhR antagonism function as a robust protective influence, preserving both metabolic homeostasis and a potent defense mechanism against osteoporosis.
