Abstract
Background, Reproductive fertility issues, influenced by environmental toxins, genetic factors, and lifestyle choices, have become a global concern. The Aryl Hydrocarbon Receptor (AhR) has garnered attention due to its involvement in male infertility mechanisms and its impact on sperm production and function. AhR, a versatile receptor expressed in various tissues, including the testes, regulates genes implicated in spermatogenesis. AhR activation has been linked to cell cycle regulation and chromatin condensation during spermatogenesis.
Objectives, This study aimed to investigate the influence of AhR activation on blood-testis barrier (BTB) integrity, focusing on the role of Tight Junction Protein 1 (Tjp1) in sperm chromatin maturity and condensation. Additionally, the study aimed to explore the effects of AhR antagonism using resveratrol (RES) and CH223191 on spermatogenesis in adult male rats.
Methods, Forty adult male rats were divided into four groups: Control, DMSO, RES, and AhR knockdown (AhR‾) group. Treatments were administered intraperitoneally over 60 days. Sperm chromatin maturity and condensation were evaluated using Toluidine Blue and Aniline Blue staining, respectively. Gene expression analysis of Tjp1 was assessed by using Rt-PCR. BTB integrity was assessed using the Evans blue test.
Result, The RES group exhibited improved sperm chromatin maturity and condensation compared to the control and DMSO groups, while the AhR‾ group showed compromised sperm chromatin integrity. Tjp1 expression was significantly upregulated in the RES group and downregulated in the AhR‾ group, indicating the potential regulatory role of Resveratrol and AhR antagonism on BTB integrity and sperm development. The Evans blue test revealed maintained BTB integrity in the RES group but impaired BTB function in the AhR‾ group.
Conclusion, Resveratrol treatment positively influenced sperm chromatin maturity, Tjp1 expression, and BTB integrity, suggesting its potential as a therapeutic agent for male reproductive health. Conversely, AhR antagonism led to compromised sperm chromatin integrity and BTB function, highlighting the critical role of AhR in spermatogenesis and BTB maintenance. These findings underscore the importance of AhR modulation in male fertility and provide insights into potential therapeutic interventions.
